We employed this modular approach for generation of a dTALE that differentiates between two highly similar DNA sequences that are both targeted by the Xanthomonas TALE, AvrBs3. These data show that this modular assembly system allows rapid generation of highly specific TALEtype DNA binding domains that target binding sites of predefined length and sequence.

DNA assembly of the yeast plasmid was tested using direct assembly in yeast (in vivo recombination), transforming the modular parts into YPH500 S. cerevisiae and quantifying assembly efficiency as the percentage of colonies that were both red and green when scanned for fluorescence.

Controlling the valency of targeting elements on nanoparticles is a major challenge. Here we describe a modular and scalable synthetic strategy to generate monovalent semiconductor quantum dots by steric exclusion. Phosphorothioate DNA is mixed with ZnS coated CdSe QDs to produce targetable and highly fluorescent nanoparticles of a single valency.

CA have previously been implemented in DNA tile assemblies (Rothemund et al. 2004; Fujibayashi et al. 2007), and it has further been suggested that it may be possible to implement CA with reactions tethered to the surface of DNA origami (Qian and Winfree 2014). In the case of DNA tile assembly.
